my first time using preseed PLEASE HELP

The directions wasn't very clear and I filled the tube all the way after doing some research and watching videos they said not to fill it up will that affect anything and how many times did you have to apply it before y'all got conceived (just curios ) how far do you stick the tube up ? I need answers and have loads of questions (THE DIRECTIONS WAS BERY UNCLEAR)
