Depends on the test.  Strips you start right after your period twice a day, clear blue digital is once a day and it tells you what cycle day to start based on your cycle length.

You don't have to start today. You would just be wasting tests. If glow is accurate, I would start testing Saturday, once a day. Then maybe twice a day starting Tuesday if still negative. I try to start testing 4-5 days before the earliest I've ovulated. Of course if it's your first time tracking, that could be hard to pinpoint. But I think starting Saturday would be fine.
