Depends on what brand you're using and what the instructions say to do. Some suggest FMU, others suggest SMU. Always always always read the insert that comes with them.

the ones that I use are the clear blue advanced digital ovulation tests and it recommends using the urine after your longest sleep until you get your high fertility reading period then it said you can test more often but need to hold your urine and not drink a lot of fluids for 4 hours. Personally I only test with my first morning urine. I would suggest reading the directions to whatever ovulation test you're using and go by then. Hopefully that helps. Good luck.
