I think the number of baby momas/daddy's doesn't really matter but I wouldn't want to get in a situation where there is a lot of drama. I think if all the dads get along with the mom and co parent peacefully for the most part and put what's best for the children first,  then I dont see a reason why your brother shouldn't date her.

I’m currently pregnant with my first but if someone was to judge me for having a baby daddy that I had to leave due to getting abused and cheated on would make me feel even worse. 
You don’t know their story but I’m saying that just make sure he wears a condom and see how the relationship goes before having kids
