Is it true that you should test first thing in the morning or does it really matter??

Answer

It doesn't really matter. The reason they say first thing in the morning is because you've been sleeping and therefore you haven't peed in several hours or had anything to drink to dilute the test. It's just easier than telling someone "Don't drink anything or pee for the next 6 hours to get the best concentration" :)
