It worked for me my first cycle using it! PCOS can definitely affect your CM because of the estrogen/testosterone dominance. I also followed the Mediterranean diet which got all of my blood work into normal limits and my ovaries free of cysts. But regarding your question I used OPKs (they worked once I got myself healthy) and when it looked like I was coming close to a positive I used 1 gram of Preseed, 2 grams on day of LH surge, and 1 gram the following day.
