You test progesterone on CD21 because ovulation should usually occur CD14 so they test progesterone when it’s at its peak at 7dpo. Depending on when you ovulate is when you test your progesterone.

Because ovulation varies woman to woman and by day 21 MOST will have ovulated. Since progesterone rises after ovulation and falls at the beginning of your period, mid way through the luteal phase is a good general rule of when to check it.
