First morning urine usually has a concentrated amount of HCG.

Mine was positive at 5pm, negstive the next day at 10 then positive again by late arvo. Same again with my 3rd pregnancy so its different for me lol

I've always tested midday or night

It will vary, depending on the amount of HCG. For me, I tested 2 or 3 days before my period was even due, about 2 in the afternoon, and I got a clear line. Some dont get a positive until they completely miss their period. 
First thing in am is when it's the most concentrated, or you can try holding it throughout the day lol.
