OPK testing

Does it matter when you take an OPK test? I just took one since my CM has thinned out quite a bit and I've had some mild cramping. The second line was very faint, so I know it's not positive yet. I'm pretty sure I haven't missed it since today was the first day I noticed the change in CM.
