When you say "DPO"...what do you mean?

For example, my fertile week ended the 27th. My most fertile day was the 25th. Am I 4 DPO (from the most fertile day), or 2 DPO (from my last fertile day of the week?

Answer

One dpo starts the day after ovulation, so the day after you get your pos OPK is your ovulation day and the next day is 1 dpo.  If you don't do OPKs then you can estimate off what glow tells you and your most fertile day, the day it says 33% chance is what they are estimating to be your O day and the day after would be 1 dpo.  O day is usually one day before fertile week is over and 1 dpo is the last day in fertile week.
