It depends on what kind of OPK you're using. If you're using a duel hormone OPK (like Clearblue digi), the instructions advise to use fmu, however, once you start getting high fertility readings, you can switch it up to testing twice a day (one in A.M., one in afternoon- after a few hours hold and limited liquid intake) as well. You can do this til you see peak results. If you're using single hormone OPK though, some women have better results NOT testing in A.M./with fmu with those (and most instructions will advise you not to). When it comes to single hormone OPK's (or the little strips), I always had better results testing around 2pm. Just make sure you've held your urine for at least 3 hours before testing & have kept liquids at a minimum. Once you see the line on the single hormone OPK's start to progressively get darker, you can also start testing 2x/day to really pinpoint O as well. As with any OPK, it also helps to track bbt at the same time to really confirm your results. Hope this helps & best of luck to you!!!

Unless your brand says to then don't use fmu! Best window is 10am - 10pm.  I always tested at 2pm and 10pm and always caught my positive! And got my bfp 4weeks ago!
