If the listing isn’t there, the job was likely filled. I’d email or call and say that I saw the listing previously but wasn’t sure if they were still hiring and explain that I was very interested in working there.

Do what you feel is right sending an email asking if there’s any open slots is fine but you’ll look better going to the building where you are trying to apply for and ask to speak with a manager/someone in charge and find out as much information as you can
